Abstract
La presente labor investigativa se llevó a efecto en la Estación Experimental del Litoral Sur (EELS) del INIAP, donde se colectaron las muestras de las especies forestales en estudio. La Estación Experimental está ubicada en el Km 26, parroquia Virgen de Fátima, Yaguachi, provincia del Guayas. La extracción de ADN se efectuó en el Laboratorio de Biotecnología de la Universidad Técnica Estatal de Quevedo ubicada en el Km. 1,5 vía Quevedo – Santo Domingo de los Tsáchilas, en la provincia de Los Ríos y el Laboratorio de Biotecnología de la EELS. Finalmente se efectuó el genotipaje de las accesiones de C.ochroxylum, M. peruiferum y M. tinctoria en el Laboratorio de Biotecnología de la Estación Experimental Santa Catalina del INIAP ubicado en la Panamericana Sur km 1, Vía Tambillo, Sector Cutuglagua, en la provincia de Pichincha. El objetivo general propuesto fue: Caracterizar molecularmente las accesiones de C. ochroxylum, M. peruiferum y M. tinctoria, para estimar el grado de diversidad genética presente en las respectivas poblaciones del Banco de Germoplasma Forestal de la Estación Experimental del Litoral Sur¬, y los objetivos específicos; Determinar el nivel de variación y relación genética existente en las accesiones de C. ochroxylum, M. peruiferum y M. tinctoria, a través de la técnica molecular RAPDs; Evaluar la eficiencia de los marcadores moleculares RAPDs para caracterizar el contenido genético de C. ochroxylum, M. peruiferum y M. tinctoria; los mencionado objetivos se sujetaron a la hipótesis; Existe variabilidad genética entre las poblaciones de C.ochroxylum, M. peruiferum y M. tinctoria detectada a través de marcadores moleculares; Los marcadores moleculares RAPDs son eficientes para la caracterización de la variabilidad genética. La técnica RAPDs, fue utilizada para evaluar las relaciones genéticas entre 75 accesiones de C.ochroxylum, 24 accesiones de M. peruiferum y 21 accesiones de M. tinctoria, las mismas que fueron colectadas en las provincias de Cotopaxi, El Oro, Esmeraldas, Guayas, Loja, Los Ríos y Manabí. El análisis estadístico “clúster análisis” y “PCO” se realizó en base al registro de polimorfismo que arrojo cada forestal como en el caso C.ochroxylum, se obtuvieron 26 bandas polimórficas a partir de 7 primers RAPDs, con M. peruiferum, se registró 49 polimorfismos utilizando 7 primers, y por ultimo con M. tinctoria se registraron 44 bandas polimórficas a partir de 6 primers. Los resultados agruparon las accesiones de cada especie entre ellas. El dendograma separó claramente grupos y subgrupos dentro de los tres forestales en estudio, en base a estos resultados, se evidencia la estructuración del material genético de las especies en estudio, además en el caso de Bálsamo se sujeta la hipótesis de denominar a los materiales colectados en la zona de la Amazonía y Catacocha como un nuevo taxón del género Myroxylon. Los resultados sugieren posibles eventos de hibridación intraespecífica dentro del género Myroxylon.

The present investigative work was took to effect in the Station Experiment of Litoral South (EELS) of the INIAP, where was collect it the sample of the trees in study. The Station Experiment, located in the Km 26, Way to Yaguachi – Guayas. The extract of DNA was make in the Laboratory of Biotecnology of the University Technique State of Quevedo, located in the Km 1,5 via Quevedo – Santo Domingo de los Colorados, and the Laboratory of the EELS. Finally was took to effect the genotyping of the samples of C.ochroxylum, M. peruiferum y M. tinctoria, in the Laboratory of Biotecnology of the Station Experiment Santa Catalina of the INIAP, located on the Panamericana South km 1, via Tambillo, Enclosure “Cutuglagua”, Pichincha. The proposed general objective was: Characterizar molecularly the accessions of C. ochroxylum, M. peruiferum y M. tinctoria, for to determinate the degree of genetic diversity in the poblations of the tree germplasm bank of the EELS, and the specific objectives; To determinate the genetic relation- variation level present in the species in study using the RAPD technique molecular; Traits the efficient the RAPD molecular markers for to characterisate the genetic content of the three trees, subject to the hypothesis; Exist genetic variability between the poblations of C.ochroxylum, M. peruiferum y M. tinctoria detected throught of molecular markers; the RAPD moleculars markers are efficient by analysing the genetic variability. The RAPD technique was used to assess the genetic relationships among 78 accessions of C.ochroxylum, 24 accessions of M. peruiferum y 21 accessions of M. tinctoria, was collected in the provinces of Azuay, Cañar, Loja, El Oro and Zamora Chinchipe Cotopaxi, El Oro, Esmeraldas, Guayas, Loja, Los Ríos y Manabí, to the Litoral of the Ecuador. The statistic analysis "cluster analysis" and "PCO" was accomplished in base to polymorphism registre how in the case C.ochroxylum able to generated 26 bands polymorphism obtained from 7 RAPD primers, with M. peruiferum, was obtain 49 polymorphism was use 7 RAPD primers, and for ultimate with M. tinctoria was obtain 44 bands polymorphism obtained from 6 RAPD primers. The results grouped the accessions of each kind between them. The dendrograma separated clearly groups and subgroups in of the three species tree in study, in base to these results, revealed the structuration of the genetic material of the species tree in study, furthermore it is supported the hypothesis of designating to the materials collected in the zone of the Amazonía and Catacocha as a new taxón of the genera Myroxylon, the result suggest possible events of hybridizing interspecific within genera Myroxylon.
